Wnt Synergist, QS11 is a cell-permeable purine compound that binds GTPase activating proteins of ADP-ribosylation factor (ARFGAPs), including AMAP1 (Kd = 364 nM) and ARFGAP1 (Kd = 620 nM) and acts as a broad specificity ARFGAP1 inhibitor. Wnt Synergist, QS11 has also been reported to block the migration of AMAP1-overexpressing MDA-MB-231 cells (86% inhibition at 2.5 μM).
Additionally, Wnt Synergist, QS11 has been shown to work synergistically with Wnt-3a to specifically activate the Wnt/β-catenin signaling pathway in vitro. A Super(8X)TOPFlash reporter assay using HEK293 cells revealed increases in basal transcription activity by application of QS11 (2-fold), Wnt-3a (40-fold), and QS11/Wnt-3a (200-fold).
