Description

Valeryl salicylate, a potent Cox-1 (cyclooxygenase-1) inhibitor, is more selective towards Cox-1 than towards Cox-2 (ovine Cox-1 IC₅₀ = 0.8 μM and ovine Cox-2 IC₅₀ = 15 mM). The inhibition of Cox-1 (PGHS-1) has been shown to take place in a time-dependent manner and is more potent than the inhibition of Cox-2 (PGHS-2). Mechanistic studies show that Valeryl salicylate acylates the active site serine of Cox-1, which prevents arachidonate binding to the Cox active site and results in suppression of Cox activity.
