8-pCPT-2′-O-Me-cAMP is an analog of cAMP which is a specific and potent membrane activator of guanine nucleotide exchange factor, Epac, but does not have any effects on PKA. 8-pCPT-2′-O-Me-cAMP can be used to stimulate the release of calcium from intracellular calcium stores and exocytosis in β cells and the insulin-secreting cell line (INS-1). Studies indicate that substitution of a 2′-methoxyl group in place of the 2′-hydroxyl group of cAMP results in the specificity of the compound towards Epac. Epac2, another guanine nucleotide exchange factor, can inhibit the effects of 8-pCPT-2′-O-Me-cAMP. Furthermore, 8-pCPT-2′-O-Me-cAMP can be useful as a pharmacological tool, analyzing PKA-independent signaling properties of cAMP.
