Description
Leptomycin B, Free Acid, a cytotixin and Streptomyces metabolite, has been shown to be a specific inhibitor of nuclear export signal-dependent transport via inhibition of CRM1. CRM1 is a protein required for nuclear export of proteins containing a nuclear export sequence (NES). Leptomycin B has been reported to block Rev-dependent export of mRNA into the cytoplasm, which inhibits Rev-dependent gene expression. Leptomycin B has also displayed a capacity to inhibit translation and export of many RNAs, including c-Fos and Cox-2 mRNAs, by inhibiting export of ribonucleoproteins. Additional experiments have shown leptomycin B to be an antibiotic which induces cell elongation and suppression of fission yeast. Furthermore, proliferation of rat fibroblasts was shown to be reversibly suppressed by leptomycin B in both the G1 and G2 phases, forcing the cells into a resting state (GO).
